在形容“数目多”的时候，“a large ...” 和 “a large number/amount of ... ” 有什么区别？

而且 a large 的原意不是 “一个大...” 吗（如 a large book/tree/area）？怎么还有数目多的意思了呢？

large在取形容词义“多数的,大量的”时,有修饰、限定可数名词或不可数名词的用法.但通常large的后面不会直接接可数名词的复数.
a large number of + 可数名词复数 a large number of people 很多人
a large amount of + 不可数名词 a large amount of money 很多钱
此题中的a large audience,重点在于名词audience,它是一个集体名词.
当集体名词audience以整体考虑时,视为单数.a large audience 大量的观众,此时它的表达形式 相当于 a + 形容词 + 名词单数
当集体名词audience的重点放在“个人”时,则视为复数
There are five audiences smoking in the theatre.有五个观众在剧场吸烟.
集体名词：指整体时,视为单数；指成员时,视为复数
常见的集体名词：audience,class,group,team,population,public等.

关于第二句话中，a large sample of，这里的large应该是规模大的意思，是大样本。
